: S-adenosyl-L-methionine + cytosine34 in tRNA precursor \rightleftharpoons S-adenosyl-L-homocysteine + 5-methylcytosine34 in tRNA precursor
The human enzyme is specific for C5-methylation of cytosine34 in tRNA precursors.
